There seem to be a lot of web comics submitting animated shorts here recently, and I think it needs to be spelled out.
Animation and Comics are two different fields.
They share many similarities, but the key differences are what make them incompatible with the other unless altered into a completely different form
Firstly, Comics rely on the Punch, Animation relies on Timing. This piece clearly shows you have not made that distinction. You see, since comics are in panels, they rely solely on the joke and only the joke. Animation, however, relies on both the timing of the joke and the delivery in real time. While the joke itself was decent and unexpected, the entire animation moved along at a somewhat slothful pace, even taking into account how short it was, and when the joke was finally delivered it really didn't have much kick. You need more visual flare, some effects. Have the scene rumble as the stampede of runners comes by, have them kick up a dust cloud. ANYTHING! It was just really plain and kind of boring. The joke, by definition, should not be be boring. Quite frankly, it should be the exact opposite.
Next, the context in comics, specifically strips, is entirely different from the context of animation. In comic strips, a lot of build up isn't needed because the dialogue of the script is often short and to the point. However in animation, just throwing someone into this world with little to no explanation of what is going on or who these characters are leaves somewhat of a emptiness to the proceedings. The reason this works in comics is because it hinges more on dialogue while animation is focused more action. Not exactly slap-stick necessarily, but subtle body language and mannerisms of speech to give us some sense of who these people are and what the fuck is going on. You seem to have gotten some of the quirks of speech down, but all the characters' faces and bodies are generic and bland. All I know is that the guy on the ground is fucked up and those three guys next to him are probably in the mafia. Choochoo, all aboard the comedy train.
Also, while somewhat subpar art may be excusable in animation due to it, well, being animation, sticks are not. Hell, sticks are even worse in comics unless you know how to pull it off and use it to your advantage, and so far the only web comics I've managed to see that can do this well are MS Paint Adventures and xkcdm, and even then that is stretching it. Sticks just make everything look sloppy and unimagined and should really never be used, ever.
